Wang and his colleagues at the KAUST Visual Computing Center, led by computer science professor Wolfgang Heidrich, have now developed a new method for quantitative phase and grayscale imaging. The device that is critical to the performance of a microscope is called a wavefront sensor. The wavefront sensor is a sensor designed for adaptive optics, which encodes the information of the wavefront phase into a grayscale image.

The team has designed an innovative high-resolution wavefront sensor that is currently being integrated into commercial microscopes to improve the performance of microscope imaging. At the same time, they also reconstructed the phase contrast image through a computer algorithm developed by themselves, and digitally retrieved the quantitative phase from a pair of images: the calibration image obtained when there is no sample and the measurement image obtained when the sample is in place.
This method simplifies the use of the microscope in several ways. Although some methods have achieved quantitative phase imaging in the past, they are costly and complex to set up and require a dedicated light source or a long time to generate the image. "Our method enables fast calculations with simple, inexpensive optics, ordinary white light sources, and real-time video transmission rates. Snapshots capture high-resolution bright-field amplitudes and accurate quantitative phase images." Heidrich said, You know, it's a combination of all these advantages. "
